Saffron Supplements: What the Research Actually Shows (And Why the Form You Take Matters)

Saffron has been used medicinally for thousands of years across Persian, Ayurvedic, and European traditions. What’s more recent is the research, and it’s more compelling than most people realise. Over the past decade, a growing body of randomised controlled trials has examined saffron’s effects on mood, emotional wellbeing, and stress response. The findings are notable. But there’s a catch: most of the clinical evidence relates to a specific, standardised extract - not the generic saffron in your spice rack.

This matters because supplement consumers deserve to understand what they’re actually buying.

 

What Saffron Is and What Makes It Active

Saffron is derived from the dried stigmas of Crocus sativus, a flowering plant cultivated primarily in Iran, Spain, and Kashmir. It takes roughly 150,000 flowers to produce one kilogram of saffron stigmas, which is part of why genuine saffron has historically been among the world’s most expensive spices.

The compounds understood to drive saffron’s biological activity are crocins (a group of carotenoid glycosides responsible for saffron’s vivid colour) and safranal (a volatile compound that contributes to its characteristic aroma). These two compound classes are the focus of most clinical saffron research.

The challenge is that raw saffron varies considerably in its concentration of these actives depending on growing conditions, harvesting, processing, and storage. A supplement that simply lists “saffron” on its label may contain highly variable amounts of the compounds the clinical research has actually studied.

 

What the Research Shows

Mood and emotional wellbeing.  Kell et al. (2017) conducted a three-arm, double-blind, randomised, placebo-controlled trial in 128 healthy adults with self-reporting low mood. Participants received either Affron® at 28 mg/day, 22 mg/day, or placebo for four weeks. At the 28 mg dose, the saffron extract significantly reduced total mood disturbance, tension, depression, confusion, and fatigue as measured by the Profile of Mood States (POMS), compared to placebo. The 22 mg dose did not produce significant improvements, indicating a dose-dependent effect.1

Adults with subclinical depressive symptoms.  Lopresti et al. (2025) conducted what is currently the largest published RCT on saffron extract, enrolling 202 adults aged 18–70 experiencing low mood but not meeting criteria for major depressive disorder. In this 12-week, double-blind, placebo-controlled trial, Affron® supplementation was associated with significantly greater improvements in depression scores on the DASS-21 compared to placebo. Approximately half of the saffron group met the study’s responder criteria, compared to approximately one-quarter in the placebo group, with separation between groups emerging by week four.2

These findings point toward saffron as a botanical ingredient worth taking seriously for mood support during periods of emotional depletion, including the kind of sustained emotional demand that characterises modern burnout.

 

Why Standardisation Matters

The clinical evidence for saffron’s effects on mood is specifically derived from Affron®, a proprietary extract standardised to a minimum concentration of bioactive crocins and safranal. This standardisation is what allows researchers to study consistent doses and draw reliable conclusions.

Generic saffron supplements, and even high-quality culinary saffron, don’t carry this guarantee. The concentration of crocins and safranal in unextracted saffron powder varies enough that a capsule from one batch may have very little in common, biochemically, with the dose used in a clinical trial.

 

A Note on Dosing and Timeline

The clinical research on Affron® has consistently used a dose of 28 mg/day. The four-week trial by Kell et al. (2017) found meaningful mood improvements within this timeframe, while Lopresti et al.’s 12-week trial tracked outcomes over a longer period. This is consistent with how botanical ingredients generally work - they tend to accumulate effect over weeks of consistent use rather than producing immediate results.

If you’re considering a saffron supplement for mood support, consistent daily intake over at least four weeks at a clinically relevant dose gives the most meaningful basis for assessing whether it’s working for you.

 

The Takeaway

The clinical evidence behind saffron as a mood-supporting botanical is more substantial than its reputation suggests. But it’s evidence is tied to a specific, standardised form of the extract. The question to ask when evaluating any saffron supplement isn’t just whether it contains saffron - it’s whether it contains the form that’s been clinically studied, at a dose that matches the research.
